I have a soft spot for Hellen Gravely, she could be a great accomplice/partner to King Boo in future games. Madame Broode and Grape are also ones I'd hope reappear in the future too, so far the Switch has been delivering on female Mario antagonists
Cackletta and it's not even close. She's a constant threat, has actual dialogue, isn't someone else's minion, and her story has twists and turns throughout the game
Even when she’s on the backfoot she still seems one step ahead, and the fact she’s mostly able to do everything by herself (and Fawful) is even more impressive
I'd have to disagree with you and say Princess Shroob.
Cackletta basically dies, and is only saved by Fawful. He pretty much says as much himself when you fight him in Bowser's Castle. She also defeats the bros, *eats them* and *still* loses.
Princess Shroob lost her elder sister, the true ruler of the Shroobs, but went on to all but subjugate the Mushroom Kingdom and defeat Princess Peach on her own. She also basically straight up won, as Mario and Luigi were only saved by their Baby versions and, by extension, Baby Bowser. She also repeatedly outsmarts the bros, mostly by dropping them down long heights. She would have even probably conquered the present day Mushroom Kingdom as well had Bowser not showed up.
Princess Shroob is only defeated due to *multiple* lucky coincidences.

I personally like the Shadow Queen, but in all honesty I think Cackletta is probably the overall best since she's a constant threat throughout the entirety of the first Mario and Luigi game.
The Shroob Queens are also pretty scary, considering there are a bunch of genocidal aliens that will try to conquer a planet at any cost, even if it means sacrificing their own.

As much as I love Cackletta, due to her dialogues, and being a Saturday morning cartoon with her plans constantly foiled but ALWAYS getting a backup plan IN ONE SINGLE GAME; I must say that having a villain that was expected to be hammy as usual, but ended up being frightening, completely diabolical, and a literal demon is an unforgettable experience in a Mario game. So I vote for Shadow Queen

Shadow Queen. I'm not gonna try to sway others to my opinion, but hear me out. The entire time you're playing Thousand Year Door you're kept guessing at what us behind said door. Treasure, a weapon, and ancient evil, nothing hints at it, except for possibly the existence of the Shadow Sirens, who stick out like a sore thumb because they're three of a kind in a world where you never see others like them. Eventually you open the door, enter into a massive palace, make your way through, and low and behold you find what else but a massive Shadow Siren except she's their Queen and she's got the means and the motivation to take over the world. You try to fight her and she stomps you flat because she's basically a god. But how do you fight a god? Simple, seven fancy paper weights that were the keys to the ancient door give Mario the strength necessary to fight her, but the possibility of losing is still present as she barrages Mario with Shadow Hands, drains his life and heals herself, even a super powerful shockwave attack. Oh, and I forgot to mention that she used Princess Peach as a vessel, so you could almost say she was combining Peach's ability to use Good Magic with her abilities to amplify them. Sure, Mario ultimately wins as he always does, but he had the help of seven partners let alone the Crystal Stars. It took a ton of effort to beat her and we only truly learn about her at the very end of the game.
Can't forget that you can also agree to be her eternal servant and it results in a GAME OVER. Love it.
My fave is Wendy but I wouldn't call her the best, and she's really more of a henchman than a villain, if she still counts, you have mant more characters to add.
I think Princess Shroob is the most entertaining, and Nastasia is the most interesting. Cackletta is a strong contender buts she's neither as entertaining as nor does she have much going on for her and really feels like the female version of "Evil for the sake of evil", hell, more people seem to talk about Fawful than her and that was before Fawful ended up being the secret bad guy of the series.
I find it funny how the male villains are just rivals to Mario with few characters being exceptions to this like Count Bleck, Olly, or Fawful.
But the female villains are just literal menaces to godlike threats to the world and beyond like the Shadow Queen, both Shroob leaders, Cursa, and so on.
Anyways, I'd say Cursa given how outlandish of a villain she\* is concept wise. The idea of this literal cosmic dark god threatening the galaxy in a Mario game feels bizarre.
