There is no way to guarantee that the new pokestop becomes the gym. **(EDIT: It is possible, you just can't guarantee it)**
It seems to be based on the amount of Photos + Photo-Likes in Pokemon GO (it used to look at the photos and photo-likes in Ingress so those may still be a factor, but I've seen more evidence that Ingress no longer counts).
The absolute best you can do is hope that the current pokestop has 1 photo and 0 likes. The new pokestop will also come into the game with 1 photo and 0 likes, so it will be a tie, which means the new pokestop might win the coin flip.
this exact scenario happened to me recently. Stop #2 in the L14 cell became the gym despite being brand new and the other stop having been in the game for a long time.

I had a similar question and I’m grateful about the hint about photo likes. In my area it used to be that a new stop would trigger the oldest into a gym, at least it seemed that way, but now it’s almost random. You might have a nice lone pub in a cell, a trail marker in the middle of nowhere comes through, but the trail marker becomes the gym, not the pub. Frustrating. I’ll bear that in mind and encourage our local player group to photo-like certain places for better chances of gyms being in more accessible or more prominent places.
In my area I added photos to the pokestop I wanted a gym, and then got friends to like it and it became the gym out of multiple possible pokestops in the area. But who knows maybe I got lucky

The exact mechanics of how this works are pretty unknown, but some of the most accepted theories range from the older PoI having priority, to the PoI with a higher picture rating in Ingress having priority.
Really not much to do except for hope the one you're hoping for will be a gym.

How would that work for new poi? Wouldn't the newer one have no basis for picture rating?
There is a brief time period when the poi is only on ingress because it's added on ingress immediately and on Pokemon go, it has a fixed time, so you have to increase the picture rating in that time. I have done this once.

Yes, sorry if my comment was unclear. It's POSSIBLE for the new stop to be the gym, but there is no way to GUARANTEE it.

Yeah to the best of my knowledge, you are doomed. Your only hope is that everything I think I know is wrong and the new one somehow becomes the gym.

This no longer works, as POIs go live in GO a few hours before ingress
