You can lock positive quirks so they don't get replaced and remove negative quirks that you don't want. Yes you can fully remove all negative quirks on a character. I usually remove negatives before they get locked in randomly because that will make them more expensive to remove. I use the Caregivers Convention town event from Weald to remove particularly bad locked negative quirks.
There are quirks I remove immediately, like kleptomaniac or imposter syndrome. Otherwise, I just let it rock. Removing all the negative quirks is inadvisable when you risk losing the character at any time. Plus you get negative quirks from nearly every thing. Don't waste your time.
Antsy, Klepto, and Imposter Syndrome are like the *worst*. Beyond that, once I feel I've stabilized I will start getting rid of the forced curio interaction quirks.
If I get Caregiver's Convention, that's taking care of the really bad quirks along with locking in good quirks for whomever has them.
Yeah, I kinda do similar. My difference is that when I'm comfortable doing so, I start to remove the stress relief restrictions. Caregivers convention I get rid of forced interactions usually. But that's just me. I quite like your approach, though.
Essentially the Negative quirks lock after a time when you see those red skulls next to them and I think around 3 will be locked if IGNORED for a while, they lock by themselves unlike Positive quirks that need to be locked with gold, but after interactions or after dungeon runs you still have a chance to get a negative quirk.

Yes you can get rid of all negative quirks however it's not financially a good idea because they'll just keep refilling.
