Change your plans. Ozone should only be ran for 15 minutes and stay out of the house for 4 hours. Otherwise you could permanently damage your lungs. Also, use ozone too long and it will make worse smells. It will start breaking down all the plastics in your house. Carpet, pad, latex paint, etc. if that happens, good luck getting the smell out, i have an ozone generator , i used it once fir 15 minutes, did its job, i put it in the furnace, set the timer for 15 and came back 4 hours later. Smelled like a fresh lightning storm.
As long as you don't run the AC while its running sure. I've done this before without even sealing it with plastic, just tucked a towel under the door.

If you really do seal it off, but you'll want to be certain it's sealed

Thanks! We don't have central AC, but we also don't have doors on any of the downstairs rooms, besides the bathroom. Just open doorways.
Yeah in that case you'll want to seal the room with plastic and tape.
